In addition to classical markers of vascular dysfunction and metabolic dysregulation, various circulating metabolites (e.g. lipoproteins, amino acids, fatty acids, and small molecules) have been associated with cognition and conversion from normal cognition to dementia. However, up until now findings have failed replication. This seminar will discuss novel and replicated findings in large well-characterized cohorts. Also, the role of genetic and environmental exposures is addressed and approaches to validate findings in models of the neurovascular unit.
